Research Abstract

ABA受容体の同定を目的とし,ビオチン化ABA(bio-ABA)を用いたファージディスプレイ法によるbio-ABA結合タンパク質の探索を試みた.まず,bio-ABAのシロイヌナズナにおける生理活性を,ABA誘導性遺伝子であるRAB18遺伝子の発現量を指標に調べたところ,ABAより低いながらもbio-ABAもRAB18遺伝子の発現を誘導できることを明らかにし,生理活性を持つことを確認した.bioABAは気孔閉鎖を誘導し、ABA様の生理作用を示すと共に、bioABAと蛍光標識アビジンを処理した孔辺細胞プロトプラストでは、細胞表層にbioABAの結合部位が観察された。bioABAを用いて,シロイヌナズナのcDNAライブラリーからスクリーニングをおこない,bio-ABA binding protein 1(BAB1)と名付けた候補遺伝子を含むいくつかの候補遺伝子を得た.予測アミノ酸配列から,BAB1はK-segmentと呼ばれるリジンリッチリピートを3つもつと考えられた.この領域は,α-helixを形成し,リガンドとの結合により構造変化がおこることが示唆されている.BAB1遺伝子にT-DNAが挿入されたbab1突然変異体を用いて,ABAによる発芽抑制と根の伸長抑制を調べたところ,いずれの表現型においてもbab1突然変異体は野生型よりもABAに対して高い感受性を示した.これらの結果は,BAB1がABAシグナルを負に制御するABA受容体として機能する可能性を示唆しており、今後の詳細な解析が期待される.
