Research Abstract

本研究の目的は、全ゲノム配列からアクチン結合蛋白質を検索し、そのアクチン結合ドメイン(ABD:Actin-binding domain)をカテゴリーに分け、それぞれのカテゴリーにあるABD遺伝子と蛍光蛋白質遺伝子の融合遺伝子を作成、細胞内で発現させアクチン繊維のみを観察するのに適し、かつ細胞毒性の無いものを選択し、生きた細胞内のアクチン繊維動態を可視化する新規プロープを開発することである。
各カテゴリーのうち、主にアクチン繊維に結合するアクチン結合蛋白質のABDと蛍光蛋白質との融合遺伝子コンストラクトを作製し、細胞内で発現させ、繊維化アクチンのみを観察するのに適したものを見いだすことを主に行なった。30種類のABDについてGFPを融合させたコンストラクトを作成し、細胞に形質導入した。それらのABDの多くは、 CHドメインを1つもしくは2つ持つもの、また、 WDドメインを複数持つものであった。そこで、 CHドメイン、 WDドメインを1つもちくは複数もつような断片とGFPとの融合蛋白質も作成した。これらのうち、アクチンの分布を示すものは、4例だけであった。また、繊維構造をみるため、全反射蛍光顕微鏡で観察したところ、その内の2つは繊維が明瞭に観察された。
単純にABDだけをGFPにつないでもアクチン繊維と結合できないものが多く見られた。たぶん、蛋白質の翻訳過程での立体形成過程に問題があるか、出来上がった蛋白質の立体構造が細胞質内で不安定なためであると思われる。ABD以外の冗長配列も加えることもおこなった。結果、いくつかの有効なアクチン繊維可視化プローブが得られた。
