Research Abstract

昨年度までの研究により、非木質試料(草本類や葉)と木本類試料では、リグニン構造分析における指標分解物(ニトロベンゼン酸化における芳香族アルデヒド類、オゾン分解におけるテトロン酸類など)の、試料あたりの収率がはっきりと異なることを明らかにすることができた。特にエリトロ型β-O-4構造のオゾン分解生成物であるエリトロン酸の細胞壁当りの収率によって、非木質試料と木本類試料をはっきりと区別することができた。一方、リグニン量と言う観点からこれら試料の差を分析しようとすると、クラーソン法を用いる限り、木本と草本の相違、堅い試料と柔らかい試料の相違を明らかにすることができなかった。このことは、草本類におけるリグニン分析にはクラーソン法が適用できないことを示しており、クラーソン法によって草本や葉などのリグニン量を評価すべきでないことを意味している。今年度は、クラーソン法の適用できないこれらの試料のリグニン量の評価のために、2つの新しい方法を適用した。一つ目は、赤外スペクトルを用いてリグニン量を評価することであり、2つ目は、昨年度申請者が開発した植物細胞壁の全溶解法をもちいてNMR的にリグニン量を評価することである。このうち、赤外スペクトルによるリグニン量の評価は、上記の指標化合物の収率ときわめて良い相関を示した。また赤外スペクトルの分析から、葉などの試料ではクチン質の存在が、クラーソンリグニン法が過大なリグニン量を与える原因の一つとなっていることが示された。非木質試料の試料調製過程において前処理として行われる各種の前抽出法が、この物質を除く上で必ずしも効果が無いことも明らかになった。
