Research Abstract

神経特異的な発現を示すアダプター分子、X11およびX11Lは動物界に広く保存されており、中央部のPIドメインおよびC末端側の2つのPDZドメインを介して、様々なタンパク質に結合し、結合タンパク質の機能や代謝を制御している。我々は、X11およびX11Lの遺伝子を二重に破壊したdouble-Ko(X11-/-X11L-/-)マウスを作成し、X11sの機能解析を進めている。X11Lは主に、興奮性の神経細胞に局在し、X11はそれ以外の抑制性神経細胞に発現していた。このため、一方の欠失は他方の補償的発現をもたらさない。X11-/-X11L-/-マウスは、突発的な側頭葉てんかん発作を示す。発作後のマウス脳を取り出し、免疫染色を行い、神経活動が高まっているc-fos陽性領域を同定した。この領域の切片を作成しcurrent-clamp法など電気生理学的解析を行い、野生型マウスに比較して興奮性能が高いことを示した。てんかんは、過興奮性で説明が可能であるため、この領域の神経細胞の興奮性に関わるチャネルなどの膜タンパク質の同定を進めている。電気生理学的解析から想定されるチャネルや受容体の制御異常にX11およびX11Lが関与している可能性を生化学的に解析中である。てんかんの多くは、発症機構が未知であるため、本研究による発作の分子機構の解明は、新たな治療薬のターゲットの開発に貢献できる。
