Research Abstract

本研究は、哺乳類の生物時計の光受容体であるメラノプシンを株化細胞や初代培養細胞に導入することにより、光照射によりサーカディアンリズムを同調できる時計細胞を構築すること、さらに、マウスへの遺伝子導入や導入細胞の移植を介し、直接光同調可能な末梢時計を作成することを目的とした。そこで、生物時計の光受容体であるメラノプシンに注目し、遺伝子導入により末梢時計を直接光感受細胞とし、光照射により体のリズムを整えることが可能かどうかを検討した。
メラノプシン強制発現プラスミドは、海外共同研究者のPanda博士より提供を受けた。導入細胞の検出のため、メラノプシン配列下流に蛍光色素GFPを挿入し、融合蛋白の強制発現ベクターを作成した。さらに、このベクターをマウス由来線維芽細胞株NIH3T3に導入し、強制発現させることに成功した。光照射は、ファイバー光源を用い、昼色光と、緑色域単色光を使用した。光受容後の細胞内情報伝達系の活性化は、メラノプシンとカルシウム結合性蛍光色素カメレオンの共導入後、FRET解析にて検討した。しかし、同一細胞である確率が必ずしも高くないこと、また、メラノプシン安定発現株へのカメレオン一過性導入は、導入効率が極端に低下することなどから、同一細胞での細胞内情報伝達の確認はできなかった。一方、昨年11月に、共同研究者のPanda博士は、強制発現株における光照射後のリズム脱同調を発表したため、同様に光照射を行ったところ、培養液の温度上昇のため、細胞に著しい障害を与えることが判明した。メラノプシンは多くの細胞に共通の細胞内情報伝達系を利用できる利点があるが、細胞内カルシウム上昇によるリスクもあり、今後はチャネルロドプシンなどの利用も考慮し、検討する必要がある。
