Research Abstract

肝臓の前がんからがん病変ならびに膵臓がんにおいて発現が亢進している,セリン/スレオニン・キナーぜ・Pim-3が,Badの112番目のセリン残基のリン酸化によってBadの不活化を起こすとともに,Bcl-XLの発現を誘導し,アポトーシスを抑制することでがん化に関与している可能性を我々は昨年度までに報告した。本年度,以下の結果を得た。
(1)肝臓、膵臓以外の内胚葉由来臓器である大腸、胃のヒトの前がんならびにがん病変でのPim-3の発現を免疫染色法にて検討した結果,大腸では腺がんの存在しない正常粘膜ではPim-3が検出されないのに対して,腺がん病変周囲の正常粘膜の約20%,アデノーマの約90%,腺がん病変の約55%で,Pim-3タンパクが検出された。同様の結果が,胃のアデノーマならびに腺がん組織の検討によっても得られた。
(2)Pim-3がBadの112番目のセリン残基を選択的にリン酸化することに基づく,Pim-3の試験管内での活性測定法を確立した。Pim-3の立体構造予測から推測された候補阻害剤14種を検討し,4種がPim-3活性を阻害し,そのうち2種が膵がん細胞株の試験管内での増殖を抑制することを認めた。
(3)ヒト膵がん細胞株でのPim-3遺伝子の恒常的発現機構を検討し,膵がん細胞株で発現が亢進している転写因子Ets-1が,Pim-3遺伝子プロモーター上のEts-1結合領域に結合し,転写を誘導していることを明らかにした。
