Research Abstract

本研究において、RECKがADAM10の内在性の阻害因子であること、RECK欠損マウス中枢神経系において、Notch受容体のリガンドであるDeltaのシェディングがADAM10依存的に亢進するために、Notchシグナルが減弱し、神経前駆体細胞の早熟分化が誘導されることを明らかにした(Nature Neurosci.10:838-45,2007)。一方、Nestin陽性細胞系列における.RECKの過剰(遷延)発現は、小脳症を引き起こした。面白いことに、このマウスでも、Notchシグナルの増強ではなく、減弱が観察された。つまり、ADAM10の活性は高すぎても低すぎてもNotchシグナルを撹乱する。実際、ADAM10欠損マウスでは、Notchシグナルが減弱するが、マウスにおいてADAM10はNotch受容体をシェディングしない。よって、後者においては、Notchリガンドと受容体を両方発現する細胞が増えるため、神経幹細胞間の方向性が生じなくなること、そして、Notchリガンドは同細胞で発現する(in-cis)受容体の活性化を抑制する作用があることが機序になると考えられた。これらの知見から、ADAM10活性が厳密に制御されることが中神経系発生において重要であることがわかった。一方、我々は、血管内皮細胞、間葉系幹細胞におけるRECKの役割を追求、RECKがNotchシグナル以外の増殖シグナルにも影響を与えることを発見した。血管内皮細胞において、RECKは、MMP依存的にインテグリン受容体の活性化を制御する。さらに、特定の血管増殖因子は、血管内皮細胞においてRECK発現を強く誘導する。RNA干渉法を用い増殖刺激下でRECKを誘導出来なくした細胞は、細胞老化を引き起こす。目下、この機序を探索中である。
