Research Abstract

系列特異的細胞消去法による免疫細胞の機能解析のシステムを立ち上げるためのモデルスタディとして、これまで機能解析がほとんど進んでいない好塩基球を標的とした好塩基球欠損マウスの樹立を目標に研究を進めた。前年度の研究で、BACクローン上で相同組み換え技術を応用して、好塩基球特異的に発現しているプロテアーゼmMCP-8をコードする遺伝子の蛋白コード領域をジフテリア毒素遺伝子あるいは緑色蛍光色素(GFP)遺伝子に置換したコンストラクトを作製した。本年度は、緑色蛍光色素遺伝子に置換したコンストラクトをマウス受精卵に導入し、BACトランスジェニックマウスの樹立を試みた。生まれてきた子マウスの尾DNAの解析から、低頻度ながら、BAC由来遺伝子が組み込まれた子マウス数匹を得た。それらのマウスの末梢血をフローサイトメトリーで解析したところ、いずれのマウスにおいてもCD49b陽性FcεRI陽性の好塩基球におけるGFPの発現が確認されなかった。そこで、mMCP-8遺伝子を含む別のBACクローンを用いて、GFP置換コンストラクトを作製しなおした。このコンストラクトをin vitroでマウス骨髄細胞に導入したところ、好塩基球特異的にGFPが発現されることが確認された。そこでこのコンストラクトをマウス受精卵に導入したところ、生まれてきた子マウスの一部で、末梢血中の好塩基球におけるGFPの発現が確認された。現在、この好塩基球GFP陽性マウスの子孫を育成中であり、これらのマウスで好塩基球特異的なGFP発現が再度確認されれば、GFPの代わりにジフテリア毒素遺伝子を組み込んだBACコンストラクトを作製して、マウス受精卵に導入する予定である。
