Research Abstract

肝がん細胞の起源となる可能性がある骨髄及び肝細胞のマーカー遺伝子を用いることによって,その動態をin vivoで探索するとともに発がんに至る分子機構の解明を目的とした。昨年度に引き続いて,マーカー遺伝子をもった骨髄細胞によって慢性肝炎モデルを構成することによって,肝細胞の再構築における骨髄細胞の関与を観察するとともに,慢性肝炎の過程で出現してくる肝がん細胞が骨髄細胞,肝細胞,または両者の融合細胞のいずれに由来するかを検討した。
1)慢性肝炎モデルの作成:Diethylnitrosamine化学発がんモデル(♂)(Lab.Invest.85:655,2005)において,放射線照射(900 cGy)したのちに同系統(C57BL/6)のGFP遺伝子導入マウス(♀)の骨髄細胞(10^7個)を移植して骨髄の再構築を行った。
(2)慢性肝炎組織の解析:門脈域を中心に骨髄細胞と肝臓由来細胞の融合が起こっていることを観察した。
(3)肝がん組織の解析:骨髄由来GFP陽性細胞は,Cytokeratin陽性の胆管由来がん細胞やAlbumin陽性の肝由来がん細胞と異なっていた。そして,骨髄由来細胞の一部はPCNA陽性の増殖能の高い細胞であるものの,がん細胞に特徴的なβ-cateninの核移行は認めず,間質を構成する細胞に分化しているものと考えられた。
これより,肝がん細胞が直接的に骨髄細胞から由来する過程は観察されなかったが,骨髄由来細胞はがん組織の間質(線維芽細胞,脈管,炎症細胞など)に分化して,発がんの微小環境に関与しでいる可能性が示唆された。
