Research Abstract

IL-27は、最近同定されたIL-6サイトカインファミリーに属するサイトカインの1つで、STAT1とSTAT3の両方を効率良く活性化し、炎症を誘導する作用と炎症を抑制する両作用を有している。IL-27は、そのレセプターであるgp130とWSX-1を介して、T細胞、B細胞やマクロファージなどの多くの細胞に作用するが、IL-27の造血幹細胞への効果は未だ報告されていない。今回、我々は、IL-27が、Stemcellfactor (SCF)と共に直接造血幹細胞に作用し、in vitroおよびin vivoにおいて造血幹細胞の初期分化誘導の制御に関与していることを明らかにした。まず、マウス造血幹細胞であるCD34^<-/low>c-Kit^-Sca-1^+Lineage marker^-(CD34^-KSL)細胞が、IL-27のレセプターであるgp130とWSX-1を共に発現していることを見出した。この造血幹細胞をIL-27とSCFでin vitroで培養すると、短期間で再構築する細胞を含むミエロイド系の造血前駆細胞が増幅し、そのいくつかは長期間の再構築活性も保持していた。次に、in vivoでの効果を調べるために、IL-27を高発現するトランスジェニックマウスを作製した。このマウスは、骨髄においてミエロイド系細胞の生成が充進し、B細胞生成が低下し、脾臓において骨髄外造血の亢進が見られた。さらに、IL-27はヒト膀帯血由来のCD34^+造血幹細胞に対しても同様に作用した。以上より、IL-27は、造血幹細胞の制御に関与する数少ないサイトカインの一つであることが明らかになった。
