Research Abstract

目的;皮膚に存在する多分化能を有する能幹細胞・メラノブラストの存在とその同定及び増殖再生機序を明らかにし、ヒト尋常性白斑患者への再生医療法の基礎を確立する。
成果および考察;1)メラノブラストの培養化:マウス新生仔の背部からトリプシン/EDTA液を使い表皮シートを単離し、さらに表皮シートから0.02%EDTAを用いてメラノブラスト細胞を分離培養化した。メラノブラストを細胞免疫学的にそのマーカーであるチロシナーゼとその関連蛋白及びメラノソーム蛋白で確認した。現状の培養法で得られる細胞数は1回に10^4個程度のため、さらなる培養条件の検討が必要である。2)ヒストン脱アセチル化酵素SIRT1とメラノブラスト:神経幹細胞ではヒストン脱アセチル化酵素SIRT1が強く発現く、さらに神経細胞への分化に必要であることを見出したが、同じ神経堤由来のメラノブラストで、SIRT1発現を検索した所、皮膚組織および初代培養のメラノブラスト、メラノーマ細胞にSIRT1が強発現していた。SIRT1の細胞内の局在を調べたところSIRT1は核ではなく、細胞質に局在していた。我々はこれまでSIRT1が核と細胞質を移動するシャトル機能を持つことを示していたが、これまで調べた培養細胞では必ず核に発現していたので、今回の発見はSIRT1の細胞質での機能を調べる上で大切であると考えられた。
意義・重要性;白斑再生医療に必要なメラノサイトの幹細胞であるメラノブラストの培養に成功した。更にメラノブラストには神経細胞に対してと同様にSIRT1が発現していた。今後、SIRT1のメラノブラストの分化における働きをSIRT1-siRNAやSIRT1の強制発現の系等を用いて検討する事により、SIRT1に作用する薬物が白斑の新規治療薬の開発に応用しえるか明らかにする事ができる。
