Research Abstract

巨細胞腫と骨肉腫の共通プロセスとして、多核巨細胞、破骨細胞の多形成という病的プロセスが存在する。本年はそれらの研究中、in vitroで破骨細胞を誘導する新たに見いだした4つの方法を詳しく解析した。一つは、骨肉腫細胞由来細胞あるいは巨細胞腫由来細胞と血液単球を、ミリポアーフィルターを介在させて、培養すると血液単球が破骨細胞へと分化するというものです。従RANKLは細胞膜上にあると考えられてきたが、この研究により、可溶性RANKLの存在が初めて証明された。この可溶性RANKLの産生機構を明らかにした。更に、血液単球を長期間(8週間以上)培養すると、何らの骨細胞誘導因子を加えなくても、破骨細胞が誘導されてくることを見いだした。ただ,培養途中でプラスチック面から血液単球を剥がすと,その後細胞が死んでしまうことから,細胞接着が重要な要因であることが分かった。三つ目は、高濃度の炭酸ガス中で血液単球を培養すると破骨細胞形成が亢進するということを初めて明らかにした。また,ヘパリン誘導骨粗鬆症の分子機構の解析を行い、ヘパリン自身が破骨細胞誘導活性を有することも初めて明らかにした。最近、CD98/FRP-1のコンディショナル・ノックアウトマウスの樹立に成功したので、この系を用い、CD98/FRP-1媒介破骨細胞形成の詳細な解析も行う予定です。本研究は巨細胞腫と骨肉腫発生病理上の共通プロセスを手がかりに、破骨細胞形成の分子機構を明らかにし、巨細胞腫や骨肉腫の病態解明の興味深い側面を解明した。更に、骨粗鬆症の予防や治療に関しても有益な情報を得ることができた。
