Research Abstract

Fibloblast Growth Factor (FGF)18はFGFファミリーに属する成長因子であり、肺、中脳の発生に重要な働きを有するとともに、骨、軟骨の形成にも関与すると報告されている。今回、歯髄組織においてもFgf18の高い発現を認めたことから、Fgf18の歯髄組織における機能について検討を行った。Fgf18欠損マウスにおいても歯の形成が認められることから歯の発生にはFgf18は関与しないが、Fgf18欠損マウス切歯歯胚はWildタイプ歯胚と比較してその大きさが小さい傾向にあった。ただ、BrdUを取り込ませて細胞増殖活性をin vivoで比較したところ、有意な差を認めるにいたらなかったことから、歯胚自体の発育が障害されたのではなく、周囲の硬組織形成の発育遅延により、歯の発育障害が惹起された可能性が高いと考えられた。なおFgf18欠損マウスは胎生致死であり、生後の歯の成長および成熟にどうFgfl8が関与するのかはいまだ不明である。歯胚形成過程におけるFgf18発現を検討したところ、Budステージより歯牙形成予定領域の上皮・間葉両組織に弱くFgf18発現をin situ hybridizationにより認め、さらにBellおよびCapステージにおいても同様に内エナメル上皮および上皮に接する間葉細胞にFgf18発現を認めた。RT-PCRにて、歯胚におけるFgf18発現は出生後増加する傾向にあることから、歯髄の成熟と関連があるものと予測される。
