Research Abstract

口腔・顎顔面の軟骨再生医療を広く普及させるため、吸収性ポリマー製造技術と中空糸製造技術を応用し、3次元増殖培養装置と非侵襲評価装置の機能を有する軟骨再生のための新規足場素材の開発を行った。吸収性ポリマー製中空糸による軟骨細胞増殖培養システムの検討としては、中空糸モジュールを用いた軟骨細胞の3次元培養において増殖状態を評価した。前年度に最適化した0.3%コラーゲンゲルを用いて中空糸モジュールでヒト軟骨細胞を増殖したところ、一回の培養で10倍増以上の良好な細胞増殖と細胞収量を得ることが出来きた。培養中、コラーゲンゲルの収縮がみられたものの、収縮分の体積で補正すると十分な細胞密度を得ることができた。吸収性ポリマー製中空糸の移植用足場素材への応用に関しては、様々な構造を有する吸収性ポリマー製の立体格子型足場素材を製造し、その力学的強度ならびに気孔率を評価した。その結果、ファイバー径1.0mm、開口径2.0mmの立体格子型足場素材がヤング率15MPa、気孔率90%と目標値に近い値を示した。本足場素材とヒト軟骨細胞を用いて再生軟骨を作製し、ヌードマウスに移植したところ、II型コラーゲンやプロテオグリカンの発現・集積が見られる良好な再生軟骨を得た。吸収性ポリマー製中空糸を用いた再生組織非侵襲評価法の開発では、吸収性ポリマーとして代表的な乳酸系ポリマー製の立体格子型足場素材を作製し、X、Y軸方向のファイバーから光源をいれて、Z軸方向のファイバーを受光ファイバーとした光学系を構築し、その光学性能を検討し、原理的な可能性を実証した。
