Research Abstract

生活習慣病、特に高脂血症などに深く関与している総コレステロール値は、現在、酵素法により計測されている。しかしながら、従来法は侵襲的操作(採血)を伴うため医療機関へ出向く必要があり、計測までに多くの工程を要するなどの問題を抱えている。そこで、本研究では皮膚コレステロールの計測に着目する一方で、分子鋳型を有する自己集合単分子膜(SAM)を用いて、簡便で非侵襲的なコレステロール計測法の開発を行った。SAMはチオール基が金に自発的に吸着することで形成されるが、分子鋳型を形成するマトリックス分子としてチオールを用い、コレステロールの分子鋳型を金電極に形成した。この分子鋳型電極は1.0mM以下のコレステロール濃度に対して直線的に変化した(相関係数:0.9983)。分子鋳型電極の感度、選択性や再現性などについて検討し、簡便で迅速な電気化学センサの開発への応用を試みた。また、皮膚コレステロールを高速液体クロマトグラフィーで計測する一方、従来の酵素法による血中濃度との比較を継続的に行った結果、良い相関が得られた。非侵襲計測法開発の観点から、皮膚コレステロールへの着目が妥当であることが示唆された。
