Research Abstract |
運動中のキネシンに負荷をかけ、そのときの構造状態を観察するための実験系を構築した。まずキネシン2量体の尾部に微小管に結合した後解離できない変異体頭部をつけることにより、運動を束縛して負荷をかけることに成功した。また一分子FRET法を用いることにより、運動中のキネシンの構造変化を検出することに成功した。これらを組み合わせることによって、キネシンの力発生の原因となる構造変化を明らかにした。
