Research Abstract

本研究は細胞性粘菌のcAMPに対する走化性の分子過程を可視化することで、細胞がゆらぎと同じレベルの濃度勾配の中で、その勾配の方向を的確に認識し、運動する方向を決定する仕組みを解明することを目的とした。その為に、タンパク質性Ca2+センサーであるカメレオンYC3.60のCa2+センシングドメインにアミノ酸変異を導入することで、これまで報告された如何なるCa2+センサーよりもCa2+に対して高い親和性(Kd=20nM)を持つ、つまり超高感度なCa2+センサーcameleon ZEROを開発した。驚いたことにこのセンサーを発現させた細胞性粘菌は野生型同様に分裂増殖することから内在のCa2+キレート効果による細胞毒性は無いと考えられた。この細胞株を用いて集合体形成時におけるカルシウム動態の観察を行ったところ、BZ(ベルーゾフ・ジャボチンスキー)反応様のスパイラルおよびターゲットパターンのみならず、時空間的にランダムに発火するカルシウム動態を観察することに成功した。
