Research Abstract

Notch受容体のEGFリピートは、特徴的なO-結合型糖鎖修飾を受け、その修飾にはO-フコース転移酵素1(OFUT1)など特異的な糖転移酵素が関与する。これまでの研究で、OFUT1には、糖転移活性の他にシャペロン活性があり、この活性がOFUT1の機能に中心的な役割を果たすこと、さらにOFUT1の作用の分子機序として、多数の連続したEGFリピートのフォルディングにOFUT1が必須な役割をすることを明らかにした。また、Notch受容体の細胞外領域における新たな翻訳後修飾として、O-HexNAc修飾の存在を明らかにした。
