精神疾患患者死後脳のDNAメチル化状態の網羅的解析

Research Abstract

二大精神疾患である統合失調症および双極性障害患者の死後脳サンプルを、米国スタンレー財団より入手し、DNAメチル化解析を行った。凍結前頭葉組織から抽出したゲノムDNAを用い、DNAメチル化状態をAffymetrix社製プロモータータイリングアレイを用いて網羅的に調べた。実験に際しては、前年度までに確立した条件を用い、神経細胞核でのみ発現している蛋白を標識し、セルソーターによる分離を行い、死後脳サンプルから神経細胞核と非神経細胞核に分離・回収した。また、それぞれの画分のDNAメチル化状態をタイリングアレイを用いてprofilingを行った。現在、主要なサンプルについてはタイリングアレイ実験を終了し、データ解析、及び、定量的PCR、pyrosequencingなどのvalidation実験を行い、疾患との関連を調べている。これらの主要な結果については現在投稿準備中である。本研究により、DNAメチル化パターンはヒト神経細胞、非神経細胞間で顕著な差異を示すことが明らかになり、本手法のように細胞群のある程度の純化操作抜きでは、ヒト脳サンプルにおけるDNAメチル化解析を行い、疾患の原因を探求することは困難であることが明らかになった。また、研究の過程で単一神経細胞のゲノム解析技術を確立することができた。現段階ではDNAメチル化解析そのものには応用できないが、本技術を基に単一神経細胞を対象としたDNAメチル化解析技術の開発が期待される。
