Research Abstract

親水性固体基板上を自発展開する脂質二分子膜に関して、自発展開駆動エネルギーを決定する要因が三つの膜一基板間相互作用(van der Waals/二重層力/水和力)あることを実験・理論計算の両面から明確化した。NaCl水溶液に関して、その濃度による相互作用変調の効果を検討した結果、濃度の増加に伴い展開速度が増加する事が明らかとなった。これは負に帯電した脂質膜と同じく負に帯電した基板との間に働く平面間相互作用が電解質濃度により変化したためと結論付けられた。本結果の定量的な妥当性は拡張DLVO理論により確認された。本機構の確認実験として、正に帯電したDOTAP二分子膜系の自発展開に対する電解質濃度依存性の実験も行なった。負に帯電した二分子膜とは逆に、電解質濃度の増加に伴い展開速度は減少した。これは本研究にて提案された平面間相互作用と自発展開ダイナミクスとの相関性を支持するものである。
次いで、金属微小構造体基板上を自発展開する二分子膜内における分子拡散性に関して、単一分子直接観察を行なった。平均二乗変位などの統計的解析の結果、構造体の配置パターンに応じて分子拡散性が変調される事があきらかとなった。最後に、金属微小構造体による分子拡散性変調に基づいた膜内での分子フィルタリング現象を発現させる事に成功した。中でも、NBD色素で標識された二つの脂質同位体を異なるフィルタ効率で分離できた事は、本システムの高い分子認識能を示すものであり、今後の微小場分子操作システムにおける新規方法論の提唱がなされたものと考えられる。
