Research Abstract

光合成初期過程における集光アンテナ超分子の高効率光エネルギー捕集・伝達機構の解明は光合成の基礎研究のみならず、超分子化学や光エネルギー変換の研究分野に大きく貢献することが期待できる。そこで、集光バクテリオクロロフィル色素の自己組織化で光機能性部位が構築されている緑色光合成細菌の集光アンテナ・クロロゾームを対象とし、単一超分子分光測定と多数を対象とした物性測定の観点からその励起状態とエネルギー移動機構の解明を目指した研究を行った。
二種類の緑色光合成細菌(Chloroflexus aurantiacusとChlorobium tepidum)から単離・精製したクロロゾームを対象として、極低温で共焦点レーザー蛍光顕微鏡による測定を行い,単一超分子レベルでの検出を行うことでひとつのクロロゾームに含まれる集光バクテリオクロロフィルの分子数を推定することに成功した。あわせて、単一クロロゾームレベルでの蛍光異方性の測定に成功し、クロロゾームの超分子構造との関連について議論を行った。
また、緑色硫黄光合成細菌のクロロゾームに含まれるバクテリオクロロフィルcとバクテリオクロロフィルeの脱金属反応の速度論解析を行い、バクテリオクロロフィルの分子構造が脱金属反応に与える影響を検討した。
