Research Abstract

平成19年度は,平成18年度に引き続き実験系の構築と改良を行った.まず,ポラライザ・アナライザ,光学干渉フィルタ,水銀ランプ,高感度CCDカメラ,その他レンズ等の光学物品を新規購入して現有の倒立型顕微鏡に組み込み,偏光観測用の実験系を構築した.平成18度に問題であった,蛍光分子と光学系では励起光の反射光および偏光度の時間スケールが小さいことによる低S/N比について,まずは,光学系にNd:YAGレーザを使用することで,励起光強度を増加させて撮影を行った.また,CCDからフォトマルチプライヤに受光系を変更した.これらにより,撮影範囲は縮小されたが,感度の向上が可能となった.その結果,蛍光分子ローダミンB等について,偏光に関する校正曲線が求められた.しかし,一方で,本研究の最終目的である2次元面の撮影と流路高さ方向面の絞込には成功していない.前者は,現手法にスキャン機能を設けるか,再度CCDカメラを用いることで解決を試みる.後者については,光学系ではなく追跡蛍光粒子・分子のサイズを大きくし,時間スケールと共に撮影空間分解能を増加させることで解決を図る.平成20年度について予算を取得しているので,引き続き測定精度の向上を目指す.
なお,H19年度に完成させた,校正実験用の薄膜温度センサとヒータについては,輸送現象に関する国際会議(ISTP-19)で発表を行う予定である.また,本手法は,蛍光粒子を用いた電場と電気浸透流速度場の2次元同時計測という斬新な計測法にも応用展開している.これらの途中結果は,国際会議microTAS2008と共に,機械学会論文集および国際学会論文集へ5月中に投稿する予定である.
