Research Abstract

遺伝情報を担うDNA分子を含むクロマチンファイバーが、細胞核内部の骨格構造とどのように相互作用しているのかを明らかにするため、その構造を試験管内で人工的に再構築し、原子間力顕微鏡による観察を行った。その結果、核内骨格の構成因子であるtopo IIタンパク質を加えることで、クロマチンファイバーがたぐり寄せられるように集合し、ループ状構造を形成することを明らかにした。また、クロマチンのアセチル化修飾を引き金に、クロマチンファイバーが弛緩して細くなり、更に、核内骨格から遊離し易い(自由度の高い)状態へと変化することを示唆する実験結果を得た。
