Research Abstract

病原性、非病原性のPseudomonas属細菌を接種したレタ、ナス、タバコでは、病徴の発現時、防御応答時の何れにもアポトーシス様PCDが誘遺される。過敏感細胸死(HR)と病徴発現時の細胞死に共通して関与するMAP kinaseに関しては遺伝子の部分配列を向定一し、細胞死に関わるタバゴのNtf6と高い相同性を示すことが明らかとなった(LsNtf6)。LsNtf6の発現は過敏感細胞死の誘導時にも発現するととから、植物の感染応答PCDに普遍的に関わる遺伝子で有ると推察された。さらに、Nicotiana benthamianaのNtf6遺伝子をノックダウンした植物ではHRのみならず、野火病の病徴発現も抑制されることが判った。また、病原性菌であるP.cichorii接種時に特異的に発現するレタス由来め遺伝子のPLATZ型Zn-finer転写因子を見出し、感受性の誘導に関わる転写制御に役割を持つことを示した。また、Ralstonia solanacearum感染時に誘導される遺伝子として、低分子熱ショックタンパク質を見出し、本遺伝子ノックダウン個体では、タバコ立枯病の発病が顕著に促進することを見いだした。機能未知ながら遺伝子のノックダウン実験から、タバコ野火病やタバコ立枯病の発病が顕著に遅延する遺伝子を見いだした。即ち、これらの細菌病の発病過程には宿主の遺伝子発現が関与していることを示している。現在、発病時に特異的に発現する遺伝子のプロモーターの単離を進めており、プロモーター活性を指標にした抵抗性/発病過程に関わる植物細胞内情報伝達系の解析を進めている。
