Research Abstract

鉄、亜鉛、銅、などの金属元素は動植物の生育にとって必須である。特に生殖成長や胚発生時の形態形成にはこれらの金属元素が重要な役割を果たすと考えられる。本研究は植物における金属栄養と生殖成長、形態形成および種子成熟との関わりを明らかにすることを目的とした。具体的には、鉄、亜鉛、銅などの金属元素と生殖成長や種子の成熟とを結ぶ物質と考えられる金属キレーター、ニコチアナミンに焦点をあて、生殖器官への金属元素の輸送システムを探るとともに、輸送後の細胞内における金属元素の制御機構を明らかにすることを目的とした。本年度はイネの生殖器官および未成熟種子への金属輸送機構の解明を中心に解析を行った。プロモーターGUS遺伝子導入イネを用いた実験により、ニコチアナミン金属錯体を輸送すると考えられるトランスポーター遺伝子ファミリーが生殖成長期および種子成熟期に発現していることおよびそれらの遺伝子発現の局在が明らかとなった。また、その他の金属イオンのトランスポーターも生殖成長期に発現していることが明らかになった。ニコチアナミンを合成するニコチアナミン合成酵素遺伝子の発現の増加および発現の局在も明らかになった。上記のトランスポーターおよびニコチアナミン合成酵素遺伝子のいずれも胚発生の初期段階から発現しており、種子成熟期には胚および背側の維管束で共通して発現がみられた。これらの結果はイネの生殖成長期および種子成熟期におけるニコチアナミンおよび金属の重要性を示唆した。
