Research Abstract |
本研究テーマでは、植物におけるカルシウム情報伝達と活性酸素生成系を介した環境応答に与える金属イオンの影響について研究を行った。その結果、オゾンや紫外線あるいは低温ショックなどの環境要因に対するタバコ細胞および植物体の応答および過敏感細胞死誘導反応においてカルシウムチャネルと二次的な活性酸素生成が重要な働きをすること、その情報伝達の過程で金属イオンがイオンチャネル等の特定の分子をターゲットに大きな影響を及ぼすことを示唆するデータを得た。具体的には、環境応答に関与する当該カルシウムチャネルが、金属カチオン(Al3+、La3+,Gd3+など)に対して強い感受性を示し、活性酸素生成は、二価鉄イオン(Fe2+)や一価の銅イオン(Cu+)の影響を大きく受けることが明らかになった。また、3系統のタバコに由来する培養細胞およびイネの培養細胞の低温応答性カルシウム情報伝達に対する16種類の希土類イオンとAl3+の影響を比較した。希土類16種類に対する感受性のタイピングとAl3+に対する強い感受性から低温応答性カルシウムチャネルが機械刺激応答性カルシウムチャネルとも活性酸素応答性カルシウムチャネルとも異なる新規のチャネルであることが明らかになった。 上記の研究から得られた知見に基づき、これを応用した研究事例として、金属イオンが直接的に植物の細胞にダメージを与えるメカニズムに着目し、植物細胞を金属ストレスから保護するためのペプチドの開発に成功している。将来的にこのペプチドを分泌する植物を作成することで、効果的な金属汚染土壌での植物の栽培やファイトリメディエーションによる環境改善に道を拓ことが期待できる。
