Research Abstract

フェニレンジアミンを代謝するBacillus cereus Pda-1および10-L-2株由来のアリルアミン N-アミノアセチルトランスフェラーゼを用いてアミン類からアセチルアミド類への変換反応の検討、同酵素の精製と特性解析、同酵素のクローニングおよび解析、大腸菌形質転換株による同酵素遺伝子の発現を行った。
同酵素の類縁酵素はこれまでに生体内での役割について詳細に解析されてきたが、物質生産を目指した研究報告はなかった。
生産特性、酵素の特性解析、形質転換株での発現などを試みた。本酵素は、単環式アミン類だけでなく多環式アミン類に対して幅広く活性を示した。
また、フェニレンジアミン類については、同化合物の2つのアミノ基のうち片方のみを位置選択的にアセチル化した。つづいて、大腸菌形質転換株にて同遺伝子を発現させ、親株由来の同酵素と基質特異性を含めて特性を比較し、安定性当は似通っていたが、基質特異性について相違を見出した。
形質転換株を利用して、アミン類からアセチルアミド類への変換反応の検討をおこなった。さらに、変換反応に要するアセチル基供与体を検索し、アセチルCoAの代わりにアセトアニリド類を代替物として利用可能であると出した。
