Research Abstract

植物は一日の周期や季節変動を予期し、各種の高次機能を統御するための時計機構を備えている。現在、シロイヌナズナにおいてはCCA1とそのホモログであるLHY、及びPRRファミリーの一つであるTOC1の3因子が、概日時計機構の中心的振動体分子であると考えられている。また、CCA1/LHYとTOC1間で形成されている正-負の転写フィードバックループにより、基本的な概日リズムが生み出されると考えられている。こうしたシロイヌナズナの時計基本機構は、光形態形成、光周性花成といった高次機能統御に重要であり、また広く高等植物に普遍的である。今回、我々はcca1/lhy/toc1三重欠損株を含む多重欠損変異株セットを分離し、これらに関して、概日リズム遺伝子発現、光周性花成、胚軸伸長といった時計関連表現型の広範な比較解析を行った。その結果、cca1/lhy/toc1植物体は概日リズムの自由継続性を消失しており、TOC1のホモログPRR9,PRR7,PRR5とともに概日時計の中心振動体分子の実体としての機能を有することが判明した。また、光形態形成に関するcca1/lhy/toc1の表現型はtoc1と同等であり、一方、花成制御に関するcca1/lhy/toc1のそれはcca1/lhyと同等であることが観察された。本結果とこれまでに蓄積されている知見から、時計機構、CDF1-CO-FTを介した花成制御、光シグナルに応答した胚軸伸長制御の各経路を統一的に理解するための遺伝学的リンクのモデルを提唱した。また、TOC1とそのホモログPRR9タンパク質の検出系を構築し、概日時計の分子実体のダイナミズムを今後解析するための基盤を整えた。
