Research Abstract

ウイルス性疾病は、細菌性疾病と異なり、抗菌剤や抗生物質では防除できないことから、有効なワクチンの開発が最も効果的なウイルス疾病対策だと考えられる。ワクチンの有効性を評価するためには、獲得免疫を評価する実験系の確立が必要であるが、魚類におけるウイルスに対するT細胞の機能は殆ど解析されていない。獲得免疫の主役であるT細胞は主要組織適合抗原(MHC)によって拘束されるため、その機能解析にはMHCの遺伝型が適合した近交系あるいはクローン系統の実験動物が不可欠である。本研究では、クローンギンブナ、それらと同系の抗原提示細胞及び標的細胞株を用いることにより、(1)T細胞のウイルス抗原特異的な増殖、(2)細胞傷害性T細胞によるウイルス感染細胞の破壊、といった獲得免疫系の応答をin vitroで再現し、魚類の抗ウイルス獲得免疫応答を評価する実験系を確立した。
