Research Abstract

日本犬の一部に見られる遺伝性赤血球GLAST欠損症の分子メカニズムの解明を目的に、不安定GLAST 分子をコードするG492S GLAST の分解機序およびGLAST 遺伝子転写活性低下機序の両者について検討した。
K562 赤芽球細胞株発現系を用いた検討からは、野生型およびG492S GLAST の分解過程に大きな差異は見られず、分解はK562 では観察できない赤芽球分化後期の成熟過程すなわち正染性赤芽球~網状赤血球に起こると考えられた。
また、GLAST 遺伝子プロモーターの活性は、上流2-3 kb の領域をMEDEP 細胞株において比較した場合に犬形質による差異が見られ、何らかの赤芽球特異的転写因子の関与が示唆された。
