Research Abstract

ラクトフェリンは骨芽細胞の分化を促進する機能を持ち、骨形成を促進するサイトカインとして再生医療への利用が期待される。このためには、有効濃度のラクトフェリンを骨芽細胞に持続的に供給するドラッグ・デリバリー・システム(DDS)の開発が必要である。〓型コラーゲンは骨組織を構成する主要な細胞外マトリックスであると同時に、様々な細胞増殖因子を内包し、徐放する性質を持つことが知られている。そこで、〓型コラーゲンが骨芽細胞分化の過程において、ラクトフェリンの徐放担体として利用可能か検討するため、ウシラクトフェリンを含有する〓型コラーゲンゲル薄膜を作成した。この薄膜上でMG63 ヒト骨肉腫由来細胞を単層培養し、デキサメサゾン添加により骨芽細胞に分化誘導すると、ラクトフェリンを含まないコラーゲンゲル薄膜と比較してアルカリフォスファターゼの活性化・オステオカルシンの産生などの分化形質の発現とマトリックスの石灰化(カルシウムの沈着)が促進された。この結果より、〓型コラーゲンゲル薄膜がラクトフェリンの徐放担体として培養骨組織の形成に有用であることが示された。
