Research Abstract

生物活性化合物の特定の原子を他の元素で置き換えることでその薬理活性を向上させる手法は、医薬品開発において重要である。特に、フッ素原子の導入により化合物の物性が大きく変化することが知られているため、選択的なフッ素化反応の開発はいろいろな応用研究に役立つと考えられる。本研究では、金属触媒を用いることでフッ素を効率的に導入する方法を開発し、その不斉触媒化の実現を目的としている。
良好な収率と選択性で反応が進行する酸性度の比較的高いジカルボニル化合物やシアノホンスホン酸エステルに対して、本年度は活性化が極めて困難なアリール酢酸誘導体の触媒的不斉モノフッ素化を検討した。パラジウム触媒では全く反応が進行しなかったが、種々検討したところ、新規ニッケル-BINAP錯体を用いる触媒系が効果的であることを見出した。興味深いことに、塩基として2,6-ルチジン、補助ルイス酸としてトリメチルシリルトリフラートを添加剤として加えた時のみ、反応がほぼ定量的に進行し、高いエナンチオ選択性(最高88%ee)でモノフッ素化体を得ることができた。得られた生成物の再結晶を行い、加水分解することで、光学的に純粋なフッ素化フェニル酢酸を得ることに成功した。この反応は、カルボン酸等価体に対する触媒的不斉フッ素化反応としては最初の例である。また、この触媒系の適用範囲を拡大する目的で、添加剤などに関して反応条件の更なる最適化を検討を行った。更に、アリール酢酸以外の基質についてもフッ素化反応を検討した。
