Research Abstract

本年度は有用な天然化合物の骨格を活かした、核内受容体の転写活性をモジュレートする新しい生理活性化合物を合成するとともに、新規なタンパク質-タンパク質相互作用をハイスループットに評価するアッセイ系の構築を達成した.すなわち大腸癌の発生に関わるWntシグナル伝達系の異常亢進を抑える有用新規化合物を探索するための、ハイスループットなアッセイ系を確立した。Wntシグナル関連タンパク質APCの変異などにより蓄積したβ-cateninは核内で転写因子TCFと結合し、標的遺伝子の転写を促進する。このβ-catenin-TCFの結合を阻害する小分子を当研究室保有の天然物ライブラリーから,見出した.具体的なアッセイはヒトβ-cateninのTCF結合領域であるアルマジロリピートを含む128〜683aa、分子量約67KDaのタンパク質を、GST融合タンパク質として大腸菌から発現し,96穴マイクロプレートに固定した。ヒトTCFタンパク質も同様に大腸菌より大量に発現させることに成功し、蛍光化TCFを作成し、β-catenin-TCF複合体をプレート上で形成させた。化合物を添加,インキュベートしたのち,過剰の化合物とTCFを除く事で,複合体形成を阻害しているかを,蛍光プレートリーダーで評価した.見出された阻害剤のうち数種は,細胞を用いたWnt シグナルのレポータージーンアッセイによる転写活性も阻害し,Wnt シグナルの阻害が,β-catenin-TCF複合体の直接的阻害であることを証明した.
