Research Abstract

まず、前年度合成した各種硫酸抱合型ステロイド標品を用い、エレクトロスプレーイオン化(ESI)-タンデム質量分析法(MS/MS)におけるフラグメントの様相を定性的観点から精査した。モデル化合物として取り上げた5種の硫酸抱合型ステロイド(デヒドロエピアンドロステロン、プレグネノロン、エストロン、テストステロン、リトコール酸の各硫酸抱合体)は、いずれも脱プロトン化分子を効率良く生成した。さらに、それらの脱プロトン化分子は、低エネルギー衝突誘起解離(CID)条件下においてそれぞれの構造に特徴的なm/z 80,96,97のイオンを生成した。このことから、硫酸エステル化は、特異性向上のみならず、ステロイドの構造推定にも有用な情報を与えることを明らかにした。
ついで、リコンビナントの硫酸転移酵素(SULT2A1)を用い、その誘導体化用機能性試薬としての可能性につき、検討を加えた。PAPS(3'-phosphoadenosine 5'-phosphosulfate)を含むリン酸ナトリウム緩衝液中、不溶性担体に固定化したSULT2A1によるデヒドロエピアンドロステロンの誘導体化に検討を加えたところ、緩和な条件下で硫酸化が定量的に進行することが判明した。さらに、固定化SULT2A1の安定性を評価したところ、4℃で保存するとき、40日後においても80%以上の活性を維持していた。また、繰り返し使用により僅かな活性の低下が認められたものの、5回使用後においても固定化直後の80%以上の活性を有することが明らかになった.
