Research Abstract

本研究の目的である、終末糖化産物(Advanced Glycation End-products;AGE)とそのレセプターであるRAGE(Receptor for AGE)のAGE結合ドメインとからなる複合体の立体構造解析及び、新規RAGEリガンドを見出すためのバーチャルスクリーニングを志向して、平成19年度は以下に示す研究をおこなった。結晶化を目的とし、安定性の向上を見込んだRAGEコンストラクトを新たに数種作成し、その大腸菌大量発現系を構築した。その結果、これまで構造解析に用いてきたコンストラクトに比べ収率が向上したものが得られた。精製した蛋白質を用い結晶化スクリーニングをおこなった。表面プラズモンセンサー(Biacore)を用いた各種RAGEとAGE等各種リガンドとの相互作用解析を行い、RAGE-リガンド相互作用を定量した。その結果、昨年度見出したリガンド以上にRAGEとの親和性が高い新規RAGEリガンドを見出すことができた。またこのリガンドと結合ドメインの相耳作用部位をNMRを用いた化学シフト摂動法及び飽和移動法で解析した。一方、BSA-AGEとRAGEとの結合が二段階反応であることを表面プラズモンセンサーによる速度論的解析から見出し、RAGEのリガンド認識機構の一端を解明した。
