Research Abstract

今年度は、胚性幹細胞における転写伸長因子S-IIの機能を明らかにするため、S-II遺伝子をホモに欠損する胚性幹細胞(ES)株の樹立を試み、これに成功した。この細胞DKO20株は野生型株と同様に増殖し、熱ショックや血清飢餓に対しても同様に応答できることがわかった。一方、S-IIと相互作用する新規転写活性化因子FESTAによる転写活性化がS-IIに依存しているか否かを解析したところ、FESTAによる転写活性化が減弱することを見出した。S-IIと相互作用しない転写因子GRIPltauによる転写活性化は、DKO20でも野生型株と同様に観察されたことから、FESTAの転写活性化の減弱は、S-IIとの相互作用に依存するものであることが明らかとなった。胚性幹細胞は試験管内で様々な細胞系譜に分化させることができる。これらの系を利用し、S-II欠損により各種細胞系譜への分化が障害されるか否かを解析することが今後の課題である。
