Research Abstract

本研究では,難治性気道炎症性疾患嚢胞性線維症(CF)気道上皮における慢性炎症分子メカニズムの解明を主目的とし,CF気道上皮細胞で高発現するTLR2遺伝子をモデル遺伝子としCF細胞におけるDNAメチル化制御機構の破綻メカニズムに関する原因解明を行った.その際,転写開始部位付近に存在するSp1結合領域(SP1-2)周辺のCpG配列(CpG#18-20)が,変異CFTR発現上皮細胞特異的に脱メチル化を受ける領域であり,また,本領域は,TLR2遺伝子の基底状態の発現制御において重要な領域であることが明らかになった.さらに,DNA脱メチル化に伴うTLR2遺伝子発現の上昇には,転写因子Sp1が重要であることを明らかにした.最後に,DNAメチル化を導入したSp1結合領域CpG DNAオリゴに対するSp1の結合性を検討したところ,本領域のメチル化は,Sp1の結合性に影響を与えなかった.一方,興味深いことに,tri-メチル化DNAオリゴに対して特異的に結合する未同定蛋白質Xが結合してくることがわかった.すなわち,CF上皮細胞におけるTLR2遺伝子発現上昇には,Sp1結合領域のDNA脱メチル化と,それに伴う未同定蛋白質Xの結合を介したSp1依存的な転写活性化が重要である可能性が示唆された.一方,non-CF上皮細胞では,Sp1結合領域のDNAメチル化が誘発されるが,このメチル化はSp1の結合には影響を与えずに,Sp1依存的な転写活性化を抑制することが示唆された.以上,本知見は,変異CFTRを発現することで発症する慢性炎症病態発症のメカニズムにおいて,SP1の重要性を説明するものである.また,本知見は,TLR2プロモーター領域においてCF関連メチル化パターン(CFSMP: CF-associated methylation pattern)を同定した極めてユニークな研究である.
