Research Abstract

本研究では、誘導型でプロスタグランジンE2(PGE2)合成の末端酵素である、膜結合型PGE2合成酵素(mPGES-1)の脳虚血障害における役割を解析した。ラット脳虚血モデルにて、mPGES-1の蛋白質、mRNAの発現誘導が認められた。またmPGES-1欠損型マウスでは、虚血後のPGE2産生が完全に消失しただけでなく、野生型マウスに比べ脳梗塞、浮腫、アポトーシス、行動障害等の脳梗塞障害が減弱した。本研究より、mPGES-1が脳梗塞障害増悪因子であることが明らかになっただけでなく、脳梗塞治療の新たなターゲットとなるものと期待される。
