Research Abstract

ポストゲノム時代と呼ばれる今日、ゲノムとタンパク質を中心とした莫大な生物学データが報告されるようになり、これら網羅的データを扱うバイオインフォマティクスが注目されている。しかしながら、今日私たちが目にする網羅的生物学研究はとかく遺伝子・タンパク質を中心に考えられがちであり、生体関連化学物質を網羅的に扱ったバイオインフォマティクス研究は未だ皆無に等しい。実際、エネルギー産生、代謝、生体系構成成分の合成・分解、生体反応系の調節・制御といった様々な生命活動には化学物質が重要な役割を果たしており、化学物質とゲノム・プロテオームとの関わり(ネットワーク)に焦点をしぼった研究の推進が、これからのポストゲノム時代のバイオインフォマティクスの一翼を担い、生命科学のさらなる発展と創薬・医療への深い貢献につながるものと予想される。そこで本研究では、ケミカルゲノミクスで集積される膨大な化合物とタンパク質との相互作用関係を情報科学的アプローチ(機械学習やパターン認識)により解析することにより、分子間相互作用ルールの包括的抽出とそれに基づく新たな分子設計手法の開発を目指してきた。具体的には、我々の独自開発手法である化合物探索計算法(相互作用マシンラーニング法)をKinaseファミリーに適用しその性能評価を行った。その結果、従来手法であるドッキング計算法や類似化合物探索手法とは比べ物にならないほどの驚異的な予測性能を示すことが証明された。
