Research Abstract

新規SAHase阻害剤の開発を目的とし、既に強力な阻害剤として知られているヌクレオシド系抗生物質ネプラノシンAの4'および5'-炭素置換誘導体の合成を行った。共通の合成中間体として、ビニルスタナンを設定した。すなわち、容易に入手可能なリボースまたはマンノースからオレフィンメタセシスを経由して調整可能なビニルスルホンに対するラジカル反応について検討した。何れの誘導体の場合にも、ラジカル条件下での脱硫的スタニル化反応を鍵として適用し、効率的に共通の合成中間体であるビニルスタナンを得た。それぞれMitsunobu反応を利用することによってプリン塩基を導入し、共通の合成中間体であるアデニルビニルスタナンへ変換した。これらに対し、パラジウム触媒を利用するクロスカップリング反応を適用することによって、フェニル、ビニル、ベンジル、エチニルなどのほぼ任意のSp2世炭素官能基を導入する事が出来た。得られた表題化合物のうち、4'-置換体について既にSAHase阻害活性を検討したが、顕著に活性を示すものは見出されなかった。しかしながら、今日まで明らかとされていなかった脱硫的スタニル化反応の機構をモデル化合物を利用することで推定機構を提唱しうる煮いたったことは、有機合成化学の領域において有用な前進と考えられる。今後5'-置換体の活性についても検討する。
