Research Abstract

ベラクトシンAは、強いプロテアソーム阻害作用を持つ天然化合物である。当該化合物の結合するプロテアソーム内のサブユニットを同定するために、細胞膜透過性の高い蛍光標識誘導体を作成した。これを用いてin vivoとin vitroの薬剤の挙動を比較したところ、in vitroでは、β5サブユニットに優先的に結合し遅い結合解離速度を示すが、in vivoにおいてはβ1サブユニットに優先的に結合し速い結合解離速度を示すことがわかった。この違いは制御サブユニットである19S複合体の影響によるものと示唆された。
