Research Abstract

糖質の消化吸収関運遺伝子であるSI、SGLT1の高糖質食摂取時における遺伝子発現の増大には,転写因子であるCdx-2もしくはCREBが関与するだけではなく,ヒストンH3ならびにH4のアセチル化が強く関与することを明らかにした。さらに,フルクトース単回投与時におけるSIならびにフルクトース輸送担体であるGLUT5の遺伝子発現増大にも,ヒストンのアセチルヒが関与することを解き明かした。さらに,GLUT5のグルココルチコイドホルモンによる遺伝子発現の増大には,グルココルチコイドホルモン核内受容体GRの203番目の脱リン酸化,211番目のリン酸化が強く関与するだけではなく,ヒストンH3の9番目のリジン残基が重要なだけではなく,ヒストンH3のりジン9番目のメチル化,ならびにアセチル化が重要であることを明らかにした。それだけではなく,小腸吸収細胞の分化時におけるSIの遺伝子発現増大,離乳期小腸におけるSIの遺伝子発現増大にも,強くヒストンH3のメチル化・アセチル化が重要であることを明らかにした。Vitamin Aの小膓における転洪に関与するタシパク質CRBPIIの出生前後の遺伝子発現の増大にも,ヒストンのアセチル化ならびに転写因子RXRならびに転写補因子CBP/p300の結合が重要であることを解き明かした。これらの結果は,小腸のおはる栄養素の消化吸取に関与する遺伝子は,本ルモン、栄養素に絶えず応答して,遺伝子の発現量を変えているだけではなく,その転写制御機構は,ヒストンのアセチル化・メチル化の変動に伴り共通の機構により調節されていることが明らかにされた。
