Research Abstract

近年、スフィンゴシン1リン酸と呼ばれる脂質メディエーターが、リンパ球の遊走、特に二次リンパ節や胸腺からの移出の過程において重要な制御分子として機能していることが明らかとなってきた。一方、呼吸器や消化器をはじめとする粘膜組織には、粘膜免疫システムと呼ばれる免疫システムが存在する。外界と常時接している粘膜免疫システムには、生体の恒常性維持を担うための特殊な細胞集団が存在するが、それら粘膜免疫システムに特有の免疫担当細胞の遊走制御におけるスフィンゴシン1リン酸の役割については、全く明らかとなっていない。本研究において、研究代表者はスフィンゴシン1リン酸受容体の阻害剤として機能することが知られているFTY720を用いることで、腸管の生体防御分子として機能している分泌型IgAの産生細胞である腹腔B細胞やパイエル板B細胞がスフィンゴシン1リン酸を遊走制御に用いていることを見いだした。さらに研究代表者は、粘膜免疫システムに特有のリンパ球である上皮細胞間リンパ球の胸腺から腸管への遊走にスフィンゴシン1リン酸を用いていることを報告した。また食物アレルギーモデルを用いた解析から、食物アレルギーの発症に関わる病原性細胞の遊走にスフィンゴシン1リン酸が関わっていること、その機能制御を行うことで新規免疫療法の開発につながることを報告した。今後これらの研究をさらに進展させることで、スフィンゴシン1リン酸を中心とした粘膜免疫制御システムの解明が進むものと期待される。
