Research Abstract

Tumor necrosis factor-related apoptosis-inducing ligand(TRAIL)は現在臨床試験が行われている有望な抗腫瘍薬である。我々は酪酸やフラボノイドなどの食品成分がTRAILの受容体であるDR5遺伝子の発現を誘導し,TRAILと併用させることでがん細胞に強力に細胞死(アポトーシス)を誘導することを発見した。TRAILも生体に存在するタンパクであるので食品成分によりTRAIL発現を誘導することが可能であると考えられる。したがって,DR5発現を誘導する食品成分とTRAIL発現を誘導する食品成分を併用させることで,がん予防効果を発揮させることが期待できる。新たに,食品添加物として用いられている植物由来のnordihydroguaiaretic acid(NDGA)がDR5の発現を誘導し,また癌細胞に対するTRAIL誘導性アポトーシスを増強することを見出した。NDGAはリポキシゲナーゼ阻害活性を持ち,リポキシゲナーゼは癌の増殖および転移を促進する酵素である。他のリポキシゲナーゼ阻害剤もDR5発現誘導活性とTRAIL感受性を増強したことから,これらの作用はリポキシゲナーゼ阻害活性を介していると考えられた。また,抗血小板薬であるdipyridamoleもDR5発現を誘導することを見出した。dipyridamoleはcaspase阻害因子であるsurvivinの発現低下も引き起こしておりDR5の発現誘導とsurvivinの発現低下を介して,癌細胞のTRAIL感受性を増強することが判明した。食品成分のひとつがNK細胞とT細胞の細胞膜上でTRAIL発現を誘導し,さらに可溶性TRAILの放出を促進することを見出した。この食品成分で処理した血球細胞はTRAILの機能に依存して癌細胞に細胞死を誘導した。現在,細胞レベルだけでなく,この食品成分を投与したマウスの生体内でTRAILの産生が増加するか,また今までに見出したDR5発現誘導物質との併用効果を検討中である。
