心不全における心筋萎縮の機序と転写因子Foxo1における転写後調節との関わり

Research Abstract

新生仔ラットより心筋細胞を単離してシャーレ上で培養し、少量のdoxorubicin(DOX,10^-7 mol/L)を添加することにより、初代培養心筋細胞は早期老化が誘導される(Aging Cell誌に発表)とともに、萎縮を起こすことを見出した。このin vitro 心筋萎縮モデルを用いて、転写因子Foxo1の発現およびリン酸化の変化についてimmunoblotting にて検討したところ、DOX添加群においてFoxolの蛋白発現量は有意な上昇を認めなかったが、Foxolのリン酸化は有意に抑制され、Foxo1蛋白の核内移行が促進されていること、Foxo1の転写活性が上昇すること、さらに筋特異的ubiquitin ligaseのAtrogin-1, Muscle-specific RING Finger-1, Ubiquitin 1igase E3〓-II蛋白の発現が有意に上昇していることを見出した。PI3-kinase/Akt経路を介して心筋萎縮を阻害する作用のあるIGF-1を添加してもこれらの変化に有意な改善効果は認められなかったが、強力な抗酸化剤であるアスコルビン酸を追加して加えると有意な改善効果がみられた。以上の結果より、酸化ストレスによりFoxo1に対する脱リン酸化酵素が誘導され、心筋萎縮が誘導されるものと考えられた(第24回日本循環器学会学術集会で発表)。この脱リン酸化酵素を同定するため、Two hybrid法によりFoxo1遺伝子をbaitとしてmouse heart muscle cDNAライブラリーをスクリーニングする実験を継続中である。
