Research Abstract

マウスβ-defensin-6過剰発現マウスにおいて、骨格筋線維の変性、再生の所見が認められた。当該マウスは、さらに、寿命の短縮、るいそう、骨密度低下、酸化ストレスの亢進など、老化の促進に相当する表現型を示した。抗菌ペプチドによる組織障害を個体レベルで明らかにした初めての研究である。抗菌ペプチドHuman β-defensin-3は、他のdefensinと比較しても、培養細胞に対して、有意に強い細胞傷害性を有した。細胞膜透過性亢進がその主要な原因と考えられ、P2受容体拮抗薬であるPPADSにより阻害された。さらに、この細胞傷害性は、抗腫瘍効果として、個体レベルでも確認された。
