Research Abstract

NPHS1は、先天性ネフローゼ症候群の原因遺伝子の一つで、NPHS1遺伝子からネフリンという蛋白がつくられる。これは、糸球体上皮細胞が作るスリット膜(血液から尿へ大きな蛋白がもれないようにしている濾過装置)の構成分子と考えられている。日本人の先天性ネフローゼ症候群患者6人が両方のNPHS1遺伝子に異常を持ち、2人が片方の遺伝子異常を持っていた。8人中4人に同じ変異が、2人に別の同じ変異があった。今回みつかったすべての遺伝子変異をそれぞれ人工的に作り、細胞の中に入れて、異常なネフリン蛋白を作らせた。その蛋白は、遺伝子変異から予想される大きさであることをネフリン蛋白に対する抗体を使って確かめた。現在、それぞれの変異蛋白と細胞の中の各器官との位置関係の違いや変異蛋白の細胞死に関係する蛋白への影響や細胞の形に関係する蛋白への影響などを調べている。
