Research Abstract

トリプシノーゲンの異所性(膵内)活性化(トリプシン生成)にひきつづいて生じる連鎖的な諸プロテアーゼの活性化によって、膵の構成細胞が自己消化されるに至るという機構が,膵炎の主要な発症機構と考えられている。我々が樹立した膵分泌性トリプシンインヒビター欠損マウスの膵腺房細胞で誘導されるオートファジー(自食作用)の役割を解析する目的で、膵臓腺房細胞で特異的にオートファジーが欠失するマウスを作製した。このマウスは生理的条件下では異常は示さないが、セルレインで膵炎刺激を誘導すると、抵抗性を示した。さらに単離した膵腺房細胞に細胞内トリプシン活性化刺激を加えると、トリプシンの活性化がほとんど検出されないことが判明した。このことは腺房細胞内トリプシン活性化、つまり、膵炎発症機構にオートファジーが重要な役割を担っていることを示している。
