Research Abstract

糖尿病末梢神経障害モデルラットの脊髄後角細胞第II層からホールセルパッチクランプ記録を行い、電気生理学的解析をした。その結果抑制性のシナプス伝達に有意差はなかったが、モデルラットではコントロールに比べてシナプス前終末からのグルタミン酸の放出が増え、興奮性が増していた。近年神経因性疼痛の治療に使用されているガバペンチンを潅流投与したところ、コントロール群では変化はないが、糖尿病群では一次求心性神経終末に作用し、興奮性神経伝達物質の放出増強を抑制することがわかった。これがガバペンチンの鎮痛作用機序の一つであると考えられた。
