Research Abstract

肥大軟骨のマーカー遺伝子であるヒトColXプロモーターをクローニングし、その断片を組み込んだルシフェラーゼ・レポーターコンストラクトの作成を行い、このレポーター遺伝子を用いて基本転写活性の評価を行った。レポーターアッセイのホスト細胞はマウステラトカルシノーマ由来未分化間葉系細胞株ATDC5を用い、エフェクターは軟骨・骨芽分化のmaster regulatorである転写因子Runx2を利用した。その結果、Runx2はヒトColXプロモーターの転写活性を著しく増強することを確認した。さらに活性を維持するための最小単位までの絞り込みを行ったところ、転写開始点の-89bpから-60bpがRunx2のCore responsive elementであることを同定した。
